INDUCCIÓN DEL METABOLISMO SECUNDARIO DE MICROORGANISMOS MEDIANTE LA ESTRATEGIA OSMAC Y LA TÉCNICA DE COCULTIVO. AISLAMIENTO Y ELUCIDACIÓN ESTRUCTURAL DE LOS METABOLITOS SECUNDARIOS PRODUCIDOS. EVALUACIÓN DE SU ACTIVIDAD BIOLÓGICA

Resumen
En la búsqueda de nuevos productos naturales bioactivos, en el presente estudio se ha investigado una selección de microorganismos (hongos y actinomicetos) fundamentalmente de origen marino, recogidos de distintas regiones geográficas, siguiendo una estrategia OSMAC (One Strain Many Compounds) para la inducción de metabolitos secundarios crípticos, utilizando diferentes medios de cultivo, inductores químicos y técnicas de cocultivo con otros microorganismos. Este enfoque permitió el aislamiento de una amplia variedad de metabolitos secundarios, tanto nuevos como conocidos, que fueron sometidos a diferentes ensayos biológicos.
Las estructuras de los nuevos compuestos se elucidaron mediante HRESIMS y RMN, así como por
comparación con la literatura, y se aplicaron métodos computacionales QM-NMR (Quantum Mechanical - Nuclear Magnetic Resonance) para determinar sus configuraciones relativas. En algunos casos, las configuraciones absolutas se asignaron mediante dicroísmo circular electrónico o por comparación de las rotaciones ópticas observadas, con las de productos naturales conocidos, relacionados estructuralmente.
In the search for new bioactive natural products, in this study a selection of microorganisms (fungi and actinomycetes), mainly of marine origin, collected from different geographical regions, was investigated following an OSMAC (One Strain Many Compounds) approach for the induction of cryptic secondary metabolites, using different culture media, chemical elicitors and co-culture techniques with other microorganisms. This approach allowed the isolation of a wide variety of secondary metabolites, both new and known, which were subjected to different biological assays. The structures of the novel compounds were elucidated by HRESIMS and NMR, as well as by comparison with the literature, and QM-NMR (Quantum Mechanical - Nuclear Magnetic Resonance) computational methods were applied to determine their relative configurations. In some cases, absolute configurations were assigned by electronic circular dichroism or by comparison of the observed optical rotations with those of known structurally related natural products.
