ANÁLISIS DE LA EXPRESIÓN Y DE LA MODULACIÓN DE LNCRNAS EN EL DESARROLLO CARDIACO

Abstract

El desarrollo del corazón es un proceso complejo que se encuentra regulado espacio-temporalmente tanto a transcripcional como post-transcripcionalmente. En este contexto hemos estudiado el papel durante el desarrollo embrionario y en la etapa adulta de lncRNAs descritos previamente en la bibliografía mostrando que estos presentan perfiles de expresión definidos y expresiones diferenciales en las distintas cámaras cardiacas. Del mismo modo hemos identificado nuevos lncRNAs cuya expresión durante el desarrollo cardiaco sugieren un importante papel en la maduración de las cámaras atriales. Asimismo hemos estudiados la posible modulación de estos por distintos factores de transcripción cardiacos, por la ruta de señalización Pitx2>Wnt>microRNAs y por factores pro-hipertroficos y pro-arritmogenicos. Estos ensayos han demostrado una modulación diferencial de los distintos lncRNAs por estos mediadores lo que sugiere papel de estos en distintas patologías. En conjunto estos datos señalan la importancia de esta clase de RNA tanto en el desarrollo como en patologías cardiacas

The development of the heart is a complex process that is regulated space-temporarily both transcriptionally and post-transcriptionally. In this context we have studied the role during the embryonic development and in the adult stage of lncRNAs previously described in the literature showing that these have defined expression profiles and differential expressions in the different cardiac chambers. In the same way, we have identified new lncRNAs whose expression during cardiac development suggests an important role in the maturation of atrial chambers. We have also studied the possible modulation of these by different cardiac transcription factors, by the signaling pathway Pitx2> Wnt> microRNAs and by pro-hypertrophic and pro-arrhythmogenic factors. These tests have demonstrated a differential modulation of the different lncRNAs by these mediators, which suggests their role in different pathologies. Together these data indicate the importance of this class of RNA in both development and cardiac pathologies
