Aislamiento y evaluación de compuestos bioactivos de origen vegetal. Contribución a la síntesis de derivados y análogos estructurales

Resumen
El objetivo de esta Tesis Doctoral fue obtener compuestos orgánicos bioactivos, tanto naturales como sintéticos, que pudieran ser de interés en los ámbitos farmacéutico y alimentario. Para ello se ha trabajado principalmente en la recuperación de compuestos de naturaleza fenólica a partir de residuos agrícolas procedentes de la poda de diversos árboles frutales, aislando y caracterizando en torno a 40 productos naturales. También se ha caracterizado químicamente un extracto comercial de arándano rojo y se han cuantificado triterpenoides activos en diversos órganos del olivo. Se ha descrito un método de aislamiento de varios fenoles bioactivos del aceite de oliva basado en el empleo de la eficaz cromatografía en contracorriente. Se ha desarrollado una síntesis ecosostenible de oleocantal y oleaceína, así como de sus derivados metilados, a escala multigramo. Se ha iniciado la síntesis de un análogo estructural simplificado de oleocantal con la finalidad última de encontrar un “cabeza de serie” con vistas al futuro. Finalmente, la evaluación de algunas propiedades biológicas de los compuestos aislados y sintetizados se ha desarrollado en colaboración con otros grupos de investigación especializados.
The purpose of this Doctoral Thesis was to obtain both natural and synthetic bioactive organic compounds that could be of interest for the pharmaceutical and food industries. For this, some work has been done mainly on the recovery of phenolic compounds from agricultural wastes, such as the pruning woods of several fruit trees, isolating and characterizing around 40 natural products. A commercial extract of cranberry has also been chemically characterized, and some bioactive triterpenoids have been quantified in various organs of the olive tree. A method for isolating various bioactive phenols from olive oil has been described based on the use of the efficient countercurrent chromatography. An eco-sustainable synthesis of oleocanthal and oleacein, as well as their methylated derivatives, has been developed on a multigram scale. The synthesis of a simplified structural analogue of oleocanthal has been initiated with the aim of finding a "lead" to be developed in the future. Finally, the evaluation of some biological properties of the isolated and synthesized compounds has been developed in collaboration with other specialized research groups.
